A number is not always what it seems to be. What is the confidence interval? How accurate and how precise is your method of assessment? Getting an accurate and precise semen count is essential in the field of Andrology and human reproduction.

The Leja slides with a chamber depth of 10 micron are especially developed for performing semen analysis with human semen. These are very well equipped for manual counting procedures.

Both slides also work very well in any CASA system. In one assessment, you will be able to perform both sperm cell concentration, motility and viscosity assessments.If the CASA system is equipped with a morphology option, this would also work very well with Leja chambers.

As all capillary filled slides, Leja slides will have a Segre-Silberberg effect (a natural occurrence when using any type of capillary filled chamber, this effect causes a constant underestimation of the sperm concentration). It means that after the assessment of the initial sperm concentration the result needs to be multiplied with a certain factor to get the true concentration.

The magnitude of this factor depends on chamber depth, dimension of the sperm cells and the viscosity of the sample.

How accurate and precise is the assessment? The Leja 2 or 4 chamber slide is perfect for a cost effective replacement of the Improved Neubauer in semen analysis according the WHO manual. It allows the assessment of viscosity (in centiPoise, cP), motility and concentration with one slide.

The same accuracy as the Improved Neubauer is achieved, but with higher precision, without additional dilutions. It saves 30 minutes of a technician's time. One slide allows the WHO method of decision making in accepting or rejecting the concentration figure.

This slide can be used for the viscosity assessment. To get a more precise concentration figure, the initial concentration figure needs to be corrected for the Segre Silberberg effect (a natural occurrence when using a capillary filled chamber; this effect causes a constant underestimation of the concentration). Only Leja slides are calibrated and a manual gives you the correction figures.

First, it can be used to determine azoospermia, and secondly it can be used to perform a post-vasectomy check. Accuracy is extremely important, this Leja slide gives you accuracy, precision, and speed!

    DETERMINATION OF AZOOSPERMIA

    Statistically speaking it is impossible to assess a concentration of zero. Therefore it seems impossible to determine azoospermia, since by definition there are no sperms. Due to the large, fixed volume of the chamber, the Leja method for determining azoospermia is the most accurate and reliable method to determine azoospermia and very low sperm count (0- 20,000/ml). The Leja 100 micron slide is the best way of getting a statistical meaningful result combined with speed.

    POST-VASECTOMY CHECK

    The Leja 100 micron chamber is more accurate than other manufactured chambers concerning the lowest level of detection and the lowest level of quantification (lowest limit of detection Makler: 100,000/ml. Leja: 40/ml; lowest limit of quantification Makler: 2,500,000/ml, Leja: 1,000/ml).

    Reliable Post-vasectomy checks are very important. Besides the fact that the patient wants to be sure the operation was successful, the doctor needs to be sure of the results in order to avoid liability claims. The benefits of wroking with the Leja 100 micron slide are that you will save a lot of time. No spinning down is needed anymore, so no samples will be rejected to a high white blood cell count. The average number of analysis needed before clearance might go down up to a factor 2!
